What happens when Gene mutation creates a non functional enzyme?
what would be the effect if a mutation in the gene for a particular aminoacyl tRNA synthetase cause the enzyme to become completely non functional?
Since aminoacyl tRNA synthetases attach amino acids to the growing end of a polypeptide chain, a defect in a particular one would mean that the amino acid it would attach could not be attached. Consequently, polypeptide synthesis would halt, or perhaps a different amino acid would be substituted because of the redundancy of the genetic code. The primary sequence of the polypeptide chain would, as a result, be altered, so when it folded into a three-dimensional conformation, its altered secondary structure — α-helices, β-pleated sheets, EF hands, etc. — would in turn give rise to a different pattern of hydrogen bonding, salt bridges, and hydrophobic interactions, altering the shape and hence the biological function of the resulting protein. If it were an enzyme, its active or allosteric sites might be changed, rendering the enzyme less efficient or perhaps useless, whereas if it was a transmembranel protein, it might not insert into the phospatidylcholine bilayer (for example).
